Defending champion Jo Pavey will miss the Great South Run on 28 October after failing to recover from a groin injury.
The Commonwealth 5000m silver medallist is back in training but does not want to risk aggravating the problem in the 10-mile event in Portsmouth.
"If it had been two weeks later she reckons she would have been on the start line," said the event's elite athletes director Matthew Turnbull.
Australian Benita Johnson also pulled out after her Chicago Marathon efforts.
Portugal's former Olympic 10,000m champion Fernanda Ribeiro and top Kenyan Rose Cheruiyot have been drafted in as replacements.
Mara Yamauchi, Liz Yelling and Tracey Morris lead the home challenge.
World marathon champion Luke Kibet heads the men's field where his strongest rival is likely to be former European 10,000m champion Chema Martinez.
